Result description
The workflow includes EVs isolation using size exclusion size exclusion chromatography and droplet digital PCR validation to evaluate let-7f-5p expression, an early marker of autoimmunity.
Small RNAs embedded in circulating EVs are extracted from 200 µL of plasma using qEV SEC columns. The RNA content is then analysed via next-generation sequencing and validated by ddPCR. let-7f-5p was found significantly upregulated in EVs from autoantibody-positive children and is enriched in CD4+ T cells, linking it to immune mechanisms in T1D.
