The IHI Call 13 is a two-stage Research and Innovation Action (RIA) under the Innovative Health Initiative Joint Undertaking (IHI JU). The call aims to accelerate healthcare innovation through collaborative public-private research addressing major health challenges. It includes three topics: (1) establishing a European healthcare innovation network, (2) developing AI-based toxicology models to reduce animal testing in drug safety studies, and (3) advancing understanding of immune-mediated diseases associated with ageing.
This topic aims to develop and validate an Artificial Intelligence (AI) Foundation Toxicology Model and a standardised weight-of-evidence framework to support regulatory decisions on waiving second-species chronic and sub-chronic toxicity studies for small molecule medicines. The project seeks to accelerate the adoption of New Approach Methodologies (NAMs), reduce reliance on animal testing, improve regulatory consistency, and enable faster, more cost-effective drug development while maintaining patient safety. The AI model should be capable of generating transparent probabilistic predictions based on large, harmonised toxicology datasets and should be designed for future extension to additional therapeutic modalities beyond small molecules.

Eligible Countries
Participation follows the standard Horizon Europe and IHI JU eligibility rules.
Funding is generally available to legal entities established in:
- EU Member States;
- Horizon Europe Associated Countries;
- other countries eligible under Horizon Europe participation rules.
Country-specific funding restrictions described in the IHI Work Programme apply where relevant.
Submission Conditions
- Research and Innovation Action (RIA).
- Two-stage submission procedure.
- Only the highest-ranked consortium from Stage 1 is normally invited to prepare a full proposal together with the pre-identified industry consortium.
- The full proposal must integrate the pre-identified industry consortium and contributing partner identified in the topic.
- Proposals must be submitted electronically through the EU Funding & Tenders Portal.
- Applicants should demonstrate expertise in AI, toxicology, pharmaceutical safety assessment, federated data management and regulatory science, together with access to appropriate datasets and high-performance computing infrastructure.
- The project should ensure continuous engagement with regulators throughout development and validation to maximise future regulatory acceptance.
Expected Outcome
Projects funded under this topic are expected to deliver:
- a validated AI Foundation Toxicology Model capable of producing transparent probabilistic predictions to support second-species waiver decisions;
- a standardised weight-of-evidence framework enabling reproducible and transparent regulatory assessments;
- practical tools, templates and training materials supporting implementation, governance, explainability and long-term maintenance of the AI model;
- increased confidence among regulators, industry and academia in AI-supported second-species waiver applications;
- a foundation for future expansion of the model to support single-species testing and additional therapeutic modalities beyond small molecules.
Scope
Applicants are expected to develop a collaborative public-private project that delivers an AI-enabled decision-support framework for pharmaceutical toxicology. The project should:
- develop and harmonise large-scale toxicology datasets from multiple organisations using secure federated data approaches;
- develop, optimise and validate an AI Foundation Toxicology Model capable of predicting whether a second-species toxicity study is likely to provide additional safety-relevant information;
- establish a transparent, reproducible weight-of-evidence framework integrating AI predictions with other toxicological evidence to support regulatory decision-making;
- validate the model through representative industry use cases and benchmarking against predefined performance metrics;
- prepare recommendations, governance structures and implementation tools to support regulatory acceptance and long-term sustainability of the model;
- engage regulators throughout the project to facilitate future regulatory adoption and potential revision of relevant international guidelines such as ICH M3(R2).
The project should build upon previous European initiatives in toxicology data sharing and New Approach Methodologies (NAMs), including projects such as BigPicture, eTRANSAFE, eTOX, imSAVAR, VICT3R and ASPIS.
